An electrochemical paper-based immunosensor for detection of specific cancer derived exosomes

Extracellular vesicles (EVs), exosomes, and microvesicles are the main mediators of intercellular communication pathways. Due to this fact that they can be detected in a variety of body solutions and are reminder of their parent cells, they have a good potential in clinical diagnoses, especially in diagnosis and prognosis of the non-invasive cancer. However, the reliable, cost-effective and fast methods are still needed to their determination. Point-of-care testing includes a wide range of diagnostic tools that achieve these goals. Electrochemical paper-based devices as cost effective, simple, portable and disposable devices with high sensitivity and selectivity can be used. In this study, a paper-based electrochemical biosensor was designed that needs only 1.5 microliters of sample volume with the detection limit of 100 exosomes per microliter and a linear range of approximately 4 times from 5×102 to 1×107 particles/µL. For fabrication of this electrochemical paper-based device, in-lab inks were synthesized, and modifiers of graphene oxide aerosol /CoFe2O4 @ chitosan nanocomposites were prepared. The characterization of nanocomposites was investigated by FE-SEM and IR techniques and their synthesis accuracy were confirmed. Isolation and purification of exosomes from MCF7 cell line was done. CD9 antibody was immobilized on the electrode for detection of exosomes. Finally, the amount of exosomes in the serum solutions was measured. The electrochemical measurment were investigated using cyclic voltametery (CV) and differential pulse voltammetry (DPV). It is suggested that this sensitive, selective and inexpensive electrochemical biosensor has a suitable potential to be easily used in miniaturized and semiautomatic devices for determination of exosomes in specific diseases.
