Methotrexate is an anti-folate and chemotherapy agent for the treatment of various cancers such as breast cancer, leukemia, lung cancer, lymphoma, and osteosarcoma [1]. It is also applied extensively for autoimmune diseases (e.g. psoriasis, rheumatoid arthritis, lupus) as an immunosuppressive and anti-inflammatory drug [2]. However, its clinical use is limited by some severe toxic side-effects such as hepatotoxicity (liver damage), ulcerative stomatitis, low white blood cell counts and even pose life threatening in the application of high-dose methotrexate [3]. Therefore, determination of methotrexate is critical in medical clinics and pharmaceutics. The interest in the simultaneous determination of methotrexate and folic acid is based on the fact that methotrexate inhibits the body’s absorption of folic acid [4]. Thus, large dose of methotrexate or long-term treatment may lead to folic acid deficiency. Methotrexate is an effective antineoplastic drug, while folic acid is a possible tumor marker for cancer screening [5]. It has been recognized that the concentration of folic acid in human blood of cancer patients becomes lower than that of healthy individuals. In addition, folic acid can prevent the toxic side effects of methotrexate. For this reason, folic acid is usually given simultaneously with methotrexate in clinical treatments [6]. Taking the above considerations, it is necessary to develop methods for the simultaneous detection of methotrexate and folic acid. Recently, due to the low cost, operational simplicity, high sensitivity, and fast response, electrochemical techniques have gained attention and have exhibited promising applications in analysis. With the research foundations in the recent years, most promising nanomaterials were extensively applied in fabrication of electrochemical sensors, with great achievements [7, 8]. In the present work, an electrochemical sensor based on screen printed graphite electrode (SPGE) modified with Fe3O4-polypyrrole (ppy)-Pd nanocomposite has been reported for determination of methotrexate. The modified SPGE showed a great enhancement effect on the oxidation of methotrexate. The prepared Fe3O4-ppy-Pd/SPGE showed linear response to methotrexate in concentration range of 0.03-100.0 M with a detection limit of 7.0 nM. Also, this sensor was applied for determination of methotrexate in the presence of folic acid. The modified SPGE decreased the overpotentials as well as resolved the overlapping of the oxidation peak potentials of methotrexate and folic acid. |
