A label-free aptasensor for detection of Cytochrome c in early stages of apoptotic cells

Cytochrome complex (Cyt c) is an important and interesting complex protein it has a heme core inside its structure between the membranes of mitochondrial where it has a role to transfer electrons [1]. In some conditions such as pathological conditions, motivating the activation of caspases and subsequent apoptotic cell death, Cyt c can be translocated out and show up in cytosol. Monitoring Cyt c can play an important role to know more about the apoptosis process in cells [2]. This work introduced a label-free aptasensor for highly sensitive detection of Cyt c, for this purpose, we used carbon nanofiber (CNF) as highly conductive material to increase the surface area of the electrode and increase the electron transfer on the surface of the electrode. In addition, graphene oxide functionalized with aspartic acid (GOAsp) was added to the electrode surface to increase the immobilization of aptamers. Its surface is coated with multiple carboxyl groups (-COOH) which makes it easier for more aptamers to immobilize on the electrode surface. We used field emission scanning electron microscopy (FE-SEM) for the investigation of the morphology of the surface. for monitoring stepwise fabrication of aptasensor employed by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV). In the present method, the label-free quantification of Cyt c is used heme core inside the structure based on the direct electron transfer between Fe+3/Fe+2-heme redox-active site selectively bound to aptamer on the glassy carbon electrode (GCE) [3]–[5]. CNF is a highly conductive material and it makes better performance of aptasensor in addition GOAsp improve immobilizing aptamers on the surface of GCE the proposed aptasensor represents a wide dynamic range from 10 nM – 100 µM with a low detection limit of 0.74 nM which is applicable for the determination of Cyt c to find apoptotic cells in early-stage.
